DescriptionPolyglutamylated tetrahydrofolate is the central cofactor of one-carbon metabolism and is biosynthesized from GTP, para-aminobenzoate (from chorismate), and glutamate in a six step reaction.[1] The first enzyme, GTP cyclohydrolase I, in the biosynthesis of tetrahydrofolate in prokaryotes, of tetrahydrobiopterin in vertebrates, and of pteridine-containing pigments in insects catalizes dihydroneopterin triphosphate from GTP. This pathway is carried out by 6 enzymes, and the complete pathway is absent in animals.
